Metabolic profiling of fruit quality variation in lemon induced by bagging cultivation based on LC-QTOF/MS technology

摘要

Abstract

[Objective]The effects of bagging treatment on lemon fruit quality and metabolic profiles were investigated using UPLC-QTOF-MS.[Method]Using Eureka lemons from Anyue County,Sichuan Province as test material,9 bagging lemons were taken as the experimental treatment group and four non-bagged lemons were taken as the control group.Conventional quality parameters including sensory traits,soluble solids content(SSC),soluble sugars,moisture content,vitamin C,total phenolics and total flavonoids were analyzed.UPLC-QTOF-MS cou-pled with untargeted metabolomics was applied to examine changes in fruit quality and metabolic accumulation post-bagging.[Result]Com-pared with the control group,bagging fruits showed no significant differences in shape or surface appearance but exhibited increased single-fruit weight,peel thickness and fruit shape.The content of soluble sugars,SSC and total flavonoids decreased,while moisture content and total phenolics significantly increased.Vitamin C content notably declined.692 metabolites between groups were identified by metabolomic analysis with 21 differentially abundant metabolites(DAMs).Among the 10 significantly up-regulated metabolites,coumarins predominated.These coumarins demonstrated positive roles in antioxidant activity,antibacterial/antipathogenic functions and plant defense signaling-enhancing lemon's antioxidant capacity,stress resistance and overall quality.KEGG enrichment analysis revealed that differential metabolites were pri-marily enriched in linoleic acid metabolism,steroid biosynthesis and biosynthesis of various plant secondary metabolites.These pathways indi-cated that bagging significantly modulated secondary metabolic processes in lemons,coordinately regulating fruit quality through stress de-fense,appearance enhancement and nutritional improvement.[Conclusion]Bagging technology effectively promotes uniform peel coloration and enhances peel glossiness in lemons.The up-regulation of coumarin metabolites in bagging fruits strengthens stress resistance,protects fruit integrity and elevates overall quality.These metabolites also exert antioxidant,anti-inflammatory and cardiovascular-protective effects in hu-mans,delivering multiple health benefits.
