疏风解毒胶囊通过调控TLRs/MyD88信号通路对急性咽炎大鼠的影响
Exploring the Effect of Shufeng Jiedu Capsules on Acute Pharyngitis Rats by Regulating the TLRs/MyD88 Signaling Pathway

Abstract
Objective:To analyze the effect and mechanism of Shufeng Jiedu Capsules in treating acute pharyngitis based on Toll-like receptors(TLRs)/MyD88 pathway.Methods:50 SD rats were randomly divided into blank group,model group,placebo group,Amoxicillin group,low-dose,medium-dose,and high-dose Shufeng Jiedu Capsules groups.The general condition changes of rats in each group were observed.The histopathological changes of rat pharyngeal mucosa tissue cells were observed under microscope.Enzyme-linked immunosorbent assay(ELISA)was used to detect the lev-els of relevant inflammatory factors,reverse transcription quantitative polymerase chain reaction(RT-qPCR)was used to detect the mRNA expression levels of TLRs and MyD88,and protein immunoblotting was used to detect the expres-sion levels of relevant factors MyD88.Results:Compared with the placebo group,ELISA results showed that the Shufeng Jiedu Capsules low-dose,medium-dose,and high-dose groups of could significantly reduce the expression levels of serum IL-1β and IgM in rats'acute pharyngitis(P<0.05),and effectively improve the expression levels of IFN-γ(P<0.05).RT-qPCR results showed that the low-dose,medium-dose,and high-dose groups of Shufeng Jiedu Capsules could inhibit the relative expression of TLR2,TLR4,MyD88 protein genes(P<0.05);Western blot results showed that the low-dose,medium-dose,and high-dose groups of Shufeng Jiedu Capsules could inhibit MyD88 protein expres-sion(P<0.05).Conclusion:Shufeng Jiedu Capsules could significantly reduce the levels of relevant inflammatory fac-tors and protein expression,and it was confirmed that this treatment program has a significant therapeutic effect on acute pharyngitis.Its mechanism of action may be to regulate the TLRs/MyD88 signaling pathway.关键词
