基于NF-κB通路探讨生肌止痛洗剂调控肛瘘术后创面修复的分子机制
Exploring the Mechanism of Action of Shengji Zhitong Lotion to Promote Postoperative Wound Repair After Anal Fistula Based on NF-κB Pathway

Abstract
Objective:Exploring the mechanism by which Shengji Zhitong Lotion regulates wound healing after anal fis-tula surgery,and also to verify whether it affects the expression of inflammation and vascular growth factor through the regulation of NF-κB signaling pathway.Methods:From July 2024 to April 2025,120 anal fistula patients who under-went surgical treatment in the Department of Anus and Intestines of Urumqi Hospital of Traditional Chinese Medicine were randomly divided into the experimental group and the control group,and the number of patients in each group was 60 cases.The experimental group was treated with the combination of muscle-generating analgesic lotion and basic treatment,and the control group was treated with the combination of potassium permanganate fumigation and basic treatment.The wound healing of the two groups was observed on the 1st,7th and 14th postoperative days.The granu-lation growth was scored.The levels of inflammatory factors TNF-α and IL-6 as well as vascular growth factor VEGF were detected by ELISA.The expression of NF-κB p65 protein was detected by WB method.Results:The wound heal-ing rate of the experimental group was significantly higher than that of the control group on days 7 and 14(P<0.05);the granulation growth score of the experimental group was significantly lower than that of the control group(P<0.05);the levels of IL-6 and TNF-α of the experimental group were significantly lower than that of the control group,whereas the level of VEGF was higher than that of the control group(P<0.05);and the expression of NF-κB p65 in the experimental group was lower than that of the control group(P<0.01)and decreased significantly with the exten-sion of the treatment time.Conclusion:Shengji Zhitong Lotion can reduce the expression of pro-inflammatory factors IL-6 and TNF-α by inhibiting the activation of NF-κB pathway,and increase the level of VEGF at the same time,so as to improve the inflammatory state of wounds,promote angiogenesis and tissue repair,and accelerate the healing process of postoperative wounds after anal fistula surgery.关键词
