基于CAPS分子标记构建蒙农S007饲用大豆DNA指纹图谱
Construction of a DNA Fingerprinting for Hybrid Forage Soybean Mengnong S007 Based on CAPS Molecular Markers

Abstract
To improve the accuracy and efficiency of forage soybean variety identification,this study focused on the newly developed cultivar Hybrid Forage Soybean Mengnong S007 from Inner Mongolia Agricultural University.The parent,wild soybean and other forage soybean varieties(lines)were used as control,and six widely cultivated soybean varieties from both domestic China and international were selected for validation.Based on whole-genome resequencing data,single nucleotide polymorphisms(SNPs)that can be cleavable by restriction endonucleases were identified to develop stable Cleaved Ampli-fied Polymorphic Sequence(CAPS)molecular markers.The stability and effectiveness of these markers were validated,and a DNA fingerprint capable of distinctly identifying S007 was constructed.The result identified four CAPS molecular markers corresponding to specific SNP loci:A T-C mutation at position 46,861,483 on soybean chromosome 6,which was cleavable by BglⅡ in both S006 and S007;a C-A mutation at position 47 583 653 on chromosome 9,which was not cleavable by BglⅡ in wild soybean or S007;a T-C mutation at position 50,331,067 on chromosome 9,which was not cleavable by BglⅡ in S003 or S007;and a C-G mutation at position 8 555 794 on chromosome 11,which was cleavable by BglII in both S007 and the soybean line Heidou 8-4-1.The resulting DNA fingerprint pattern for Mengnong S007 forage soy-bean was"+--+(-)",demonstrating high variety specificity.The DNA fingerprint constructed in this study by CAPS markers enabled complete discrimination of all nine soybean varieties,including S007,providing an efficient,accurate,and cost-effective method for variety identification.This research pro-vides a reliable technical and theoretical reference for the examination,registration,and popularization of new forage soybean varieties,as well as for constructing fingerprint profiles in other plant species.关键词
