Preliminary report on the study of the inhibitory effect of heat treatment on yellow vein clearing virus in isolated lemon stem segments

Abstract
Heat treatment is an important method in the research of virus elimination,resistance and tolerance of lemon.In vitro culture can overcome environmental influences,shorten the breeding cycle and improve the propagation coefficient.Experiments on the temperature and duration of heat treatment were carried out on the stem segments of Eureka lemon under in vitro condi-tions,and citrus yellow vein clearing virus(CYVCV),the most harmful virus to Eureka lemon,was taken as the research object to detect the virus-inhibiting effect of heat treatment by real-time quantitative PCR.The results showed that with the increase of heat treatment temperature,the tissue structure of in vitro stem segments was damaged to varying degrees and the electrical conductiv-ity was increased.The in vitro stem segments treated at 51℃ for 30 min could develop normally with the viral load significantly inhibited,which was the extreme tolerance condition of heat treatment for in vitro Eureka lemon stem segments.关键词
