BAFF基因多态性与福建汉族人群系统性红斑狼疮易感性及中医证型相关性
Association of BAFF Gene Polymorphisms with Susceptibility to Systemic Lupus Erythematosus and Chinese Medicine Syndrome Patterns in Han Chinese Population in Fujian Province

Abstract
Objective To study the polymorphisms of B-cell activating factor(BAFF)promoter-871C/T gene and the susceptibility of systemic lupus erythematosus(SLE)patients in Fujian Han population and its correlation with different Chinese Medicine(CM)patterns of SLE.Methods A total of 420 SLE patients met the inclusion criteria.CM syndrome differentiation types were classified.Additionally,70 healthy Han Chinese individuals from Fujian undergoing routine health examinations were enrolled as the healthy control group.Next-generation sequencing technology was employed to detect genetic polymorphisms at the BAFF gene rs9514828 locus in both SLE patients and healthy controls.x2 test was used to compare the BAFF gene rs9514828 genotype and allele frequencies among different CM syndrome groups in SLE patients and to analyze differences with healthy controls.Logistic regression analysis was employed to examine the association between BAFF gene rs9514828 genotype and allele variants with SLE susceptibility and different CM syndromes.Results(1)Among SLE patients,the most common CM pattern differentiation was Turbid Toxin Obstruction Pattern(92 cases).There were statistically significant differences in the overall distributions of disease activity index(SLEDAI)scores,blood sedimentation rate(ESR),and complement C3 in the six groups of SLE patients with different Chinese medicine certificates in Fujian Han(P<0.05,P<0.01).(2)Three genotypes(CC,CT,TT)were detected at the BAFF gene rs9514828 locus in Fujian Han Chinese SLE patients.The CT genotype frequency was higher in the SLE group than in healthy controls,while the CC genotype frequency was lower(P<0.05).The results of Logistic regression analysis suggested that the Logistic regression coefficient of CT genotype at the rs9514828 locus of the BAFF gene in the SLE group was 0.788,and the OR and 95%confidence interval were 2.177(1.211~3.912),respectively(P<0.05),compared with that of the healthy control group.(3)The differences in genotype and allele distribution at the BAFF rs9514828 locus in Fujian Han patients with six different CM syndrome types of SLE were statistically significant(P<0.05).The results of Logistic regression analysis suggested that the Logistic regression coefficients of CT genotypes of SLE Fierce Heat Toxin Pattern,Turbid Toxin Obstruction Pattern,and Gan Qi Stagnation with Toxin Accumulation Pattern BAFF gene rs9514828 locus were 1.351,0.828,and 0.798,respectively,and the ORs and 95%confidence intervals were 3.861(1.722~8.661),2.290(1.132~4.632),and 2.222(1.010~4.886),respectively,as compared with those of the healthy control group(all P<0.05).Compared with the healthy control group,the Logistic regression coefficients of the T allele of SLE Fierce Heat Toxin Pattern and Heat Toxin Burning Yin Pattern were 0.751 and 0.603,respectively,and the OR and their 95%confidence intervals were 2.119(1.265~3.548)and 1.828(1.084~3.083),respectively(all P<0.05).Conclusions Turbid Toxin Obstruction Pattern was dominant in Chinese Han SLE patients in Fujian.BAFF rs9514828 locus might be a susceptibility gene for SLE in Fujian Han Chinese populations,with CT genotype potentially increasing SLE risk.The CT genotype might possibly elevate susceptibility to Fierce Heat Toxin Pattern,Turbid Toxin Obstruction Pattern,and Gan Qi Stagnation with Toxin Accumulation Pattern.Carrying T allele might increase risk for Fierce Heat Toxin Pattern and Heat Toxin Burning Yin Pattern.关键词
