摘要

Abstract

D-dimer detection is a crucial step in diagnosis of venous thromboembolism(VTE).In this study,a typical"sandwich"lateral flow immunoassay(LFIA)was established via modification of AuPd@FexOy nanoenzyme(AuPd@FexOy)with the specific antibodies of D-dimer.The AuPd@FexOy NP-labeled LFIA(AuPd@FexOy-LFIA)was used for rapid detection of D-dimer levels in human plasma.Due to its excellent peroxidase-like(POD-like)activity,in the enhanced colorimetric mode of AuPd@FexOy catalyzing H2O2 oxidation of 3,3',5,5'-tetramethylbenzidine(TMB),the limit of detection of AuPd@FexOy-LFIA was as low as 2.4 ng/mL,which was significantly lower than the health threshold of 500 ng/mL.Furthermore,the AuPd@FexOy-LFIA had good selectivity,reproducibility,and storage stability.The D-dimer detection results of 41 clinical plasma samples were well correlated with those of commercially available enzyme-linked immunosorbent assay(ELISA)detection kits(R2=0.98).The research results indicated that the AuPd@FexOy-LFIA could be used for rapid(<15 min)and sensitive detection of D-dimer in plasma,which showed potential in clinical diagnosis.
