摘要

Abstract

Objective To investigate the regulatory mechanism of miRNA-LOXL1 in oxidative stress in lens epi-thelial cells.Methods A prospective case-control study.Using H2O2 induced oxidative stress to treat human lens epithe-lial cells(SRA01/04 cell line),mRNA and miRNA correlation analysis was conducted through a model of silencing LOXL1 gene,and enrichment analysis was performed on significantly different miRNA target genes.Results mRNA analysis:There were 445 up-regulated differentially expressed genes and 568 down-regulated differentially expressed genes between the ssi LOXL1 group and the ssiNC group.There were 2 791 up-regulated differentially expressed genes and 1 375 down-regulated differentially expressed genes between the ssi LOXL1+H2O2 group and the ssiNC+H2O2 group.There were 214 up-regulated differentially expressed genes and 84 down-regulated differentially expressed genes in the ssiNC+H2O2 group compared to the ssiNC group.The ssiLOXL1+H2O2 group showed 2 703 up-regulated differentially expressed genes and 986 down-regulated differentially expressed genes compared to the the ss LOXL1 group.miRNA analysis:There were 81 up-regulated differentially expressed genes and 104 down-regulated differentially expressed genes between the ssi-LOXL1 group and the ssiNC group.There were 170 up-regulated differentially expressed genes and 191 down-regulated dif-ferentially expressed genes between the ssi LOXL1+H2O2 group and the ssiNC+H2O2 group.There were 61 up-regulated dif-ferentially expressed genes and 51 down-regulated differentially expressed genes in the ssiNC+H2O2 group compared to the ssiNC group.The ssiLOXL1+H2O2 group showed 120 up-regulated differentially expressed genes and 132 down-regulated differentially expressed genes compared to the ss LOXL1 group.The LOXL1 gene can affect the gene expression of oxidative stress lens cell lines.Conclusions The LOXL1 gene plays an important role in oxidative stress of lens epithelial cells through PI3K Akt signaling,TNF signaling pathway,JAK-STAT signaling pathway,cell adhesion molecule,MAPK sig-naling pathway,etc.
