基于lncRNA-MIAT/miR328a-5p/LPL轴探讨羊藿三七方干预2型糖尿病性勃起功能障碍大鼠的机制
Mechanism of Yanghuo Sanqi Formula on Type 2 Diabetes Mellitus Erectile Dysfunction Model Rats Based on the lncRNA MIAT/miR328a-5p/LPL Axis

Abstract
Objective To explore the mechanism by which Yanghuo Sanqi Formula(YHF)intervenes in erectile dysfunction in type 2 diabetic(T2DMED)rats via the lncRNA-MIAT/miR328a-5p/LPL axis.Methods A T2DMED rat model was induced using streptozotocin(STZ)combined with a high-fat diet.After modeling,SD rats were divided into a model group,a tadalafil group,YHF groups(low,medium,and high doses),and an lncRNA-MIAT inhibitor group,with a blank group set up as well,with 6 rats in each group.The YHF groups were administered low,medium,and high doses of YHF aqueous solution via gavage.The tadalafil control group was administered tadalafil aqueous solution via gavage.The lncRNA-MIAT inhibitor group was administered physiological saline via gavage and lncRNA-MIAT inhibitor injection(50 μL/qw).The other two groups were administered physiological saline via gavage.After 28 days of intervention,the following parameters were determined:intracavernosal pressure(ICP),mean arterial pressure(MAP),the histopathology of corpus cavernosum tissue,related signaling axis sequences,as well as interleukin-1β(IL-1β),interleukin-6(IL-6),endothelial nitric oxide synthase(eNOS),and nitric oxide(NO)content.Results Compared with the blank control group,the model group exhibited increased IL-1β and IL-6 levels,as well as decreased ICP and ICP/MAP(P<0.001),while MAP was not significantly different between the two groups(P>0.05).Compared with the model group,the high-dose YHF group exhibited significantly increased ICP,increased ICP/MAP,lncRNA-MIAT,lipoprotein lipase(LPL),and NF-κB levels,NO,glutathione(GSH),and eNOS,as well as decreased miR-328a-5p.The medium and high-dose YHF groups exhibited reduced IL-1β and IL-6 levels(P<0.05).Conclusion YHF alleviates endothelial dysfunction and treats T2DMED rats via regulating the lncRNA-MIAT/miR328a-5p/LPL signaling axis to reduce oxidative stress and enhance eNOS enzyme activity.关键词
