黑果枸杞+肉苁蓉调节PI3K/Akt信号通路对老年性骨质疏松大鼠影响的研究
Research on the effect of Fructus Lycii Ruthenic and Herba Cistanches on senile osteoporosis in aged rats via regulation of the PI3K/Akt signaling pathway

Abstract
Objective To investigate the effect of the medicinal couplet of Fructus Lycii Ruthenic and Herba Cistanches(FLRHC)on senile osteoporosis(SOP)in aged rats through regulating the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)signaling pathway.Methods Forty-eight SD rats were randomly divided into a blank group,an aging model group,a vitamin E(VE)group(0.018 g·kg⁻1),a PI3K activator(740 Y-P)group(10 mg·kg⁻1),a FLRHC group(1440 mg·kg⁻1),and a FLRHC+740 Y-P group.Micro-CT was used to analyze bone microstructure.HE staining was performed to observe femoral histopathology.Alkaline phosphatase calcium-cobalt staining and tartrate-resistant acid phosphatase(TRAP)staining were used to detect osteoblast and osteoclast activity,respectively.Serum levels of tumor necrosis factor-α(TNF-α),bone alkaline phosphatase(BALP),and tartrate-resistant acid phos-phatase 5b(TRACP-5b)were measured by ELISA.The protein expression of PI3K and Akt in femoral tissue was determined by West-ern blot(WB).Results Compared with the blank group,rats in the aging model group exhibited impaired femoral microstructure,as evi-denced by significantly decreased bone mineral density(BMD),bone volume/total volume(BV/TV),trabecular number(Tb.N),and trabecular thickness(Tb.Th),along with increased trabecular separation(Tb.Sp)and structure model index(SMI)(all P<0.05).Pathological bone damage was obvious,serum TNF-α and TRACP-5b levels were elevated,BALP level was reduced,and the protein expression of PI3K and Akt was increased(all P<0.05).Compared with the aging model group,the FLRHC treatment ameliorated bone microstructure,increased BMD,BV/TV,Tb.N,and Tb.Th,decreased Tb.Sp and SMI,alleviated pathological damage,modulated the serum biomarkers,and down-regulated the expression of PI3K and Akt proteins(all P<0.05).However,the effects of FLRHC were significantly antagonized by co-administration with the PI3K activator 740 Y-P.Conclusion The medicinal couplet of FLRHC may alle-viate senile osteoporosis in aged rats,potentially through regulating the PI3K/Akt signaling pathway.关键词
