摘要

Abstract

Objective:To investigate the influence of MICAL2PVs on the migration and proliferation of colorec-tal cancer cells(CRC).Methods:Firstly,RT-PCR,RT-qPCR,and Western blot were employed to detect the expres-sion of MICAL2PVs in CRC cell lines and tissues.After overexpressing or silencing MICAL2PVs in CRC cells,the effects of MICAL2PVs on cell migration were explored using wound healing assay and Transwell assay;the impact of MICAL2PVs on cell proliferation was determined through MTT assay and soft agar colony formation assay;the expression level of EMT-related markers in the cells were detected by Western blot.Results:The CRC cells and tissues only expressed MICAL2PVa and MICAL2PVb.Moreover,compared with the adjacent non-cancerous tissues,the expression levels of MICAL2PVs were significantly reduced in the CRC tissues(P<0.05).In comparison with the control cell line,the HCT8 stable cell line with silenced MICAL2PVs exhibited increased cell migration rate,a higher number of migrating cells,and sig-nificantly increased clone formation and cell viability(all P<0.05).When MICAL2PVb was overexpressed,the protein level of E-cadherin increased,while that of Vimentin decreased in CRC cells;conversely,when MICAL2PVs were silenced,the protein level of E-cadherin decreased,while that of Vimentin increased in CRC cells.Conclusion:MICAL2PVs can inhibit the migration and proliferation of CRC cells and are involved in regulating EMT.
