Abstract
Polygonum cuspidatum was used as the raw material to optimize the extraction and purification process for high-purity resveratrol.Different extraction methods,column chromatography resin types,ethanol concentrations for column elution,and ethanol concentrations for crystallization were systematically investigated.The skin-whitening efficacy of the purified resveratrol was evaluated using tyrosinase inhibition assays and zebrafish melanogenesis inhibition tests.The results demonstrated that the optimal conditions for the water-ethanol extraction and purification of high-content resveratrol were as follows:enzymatic hydrolysis-assisted stirring extraction as the extraction method,LXP-6 resin for column chromatography,70%ethanol as the column eluent,and 40%ethanol as the crystallization solvent.Under these optimized conditions,the average purity of resveratrol reached 99.13%.Evaluation of skin-whitening efficacy indicated that resveratrol strongly inhibited tyrosinase,with an IC₅₀ value of 8.58 μg/mL,markedly lower than kojic acid(19.08 μg/mL).In addition,treatment with 50 μg/mL resveratrol extract inhibited melanin production in zebrafish embryos by 23.76%,showing a statistically significant difference compared with the blank control group(P<0.05).These results indicate that resveratrol possesses potential skin-whitening effects through both tyrosinase inhibition and melanogenesis suppression pathways.关键词
