番茄褪绿病毒侵染茄子的分子鉴定
Molecular identification of tomato chlorotic virus infecting eggplant
摘要
Abstract
[Objective]Tomato chlorosis virus(ToCV)can infect solanaceous vegetables such as tomato,pepper and eggplant,resulting in a decrease in yield and quality.This project aims to find out the types of eggplant virus diseases collected from the greenhouse vegetable planting area in Yingxia Township,Korla City,Xinjiang in the hope of laying a theoretical foundation for the prevention and control of the disease.[Methods]The specific primers of ToCV,TSWV,TMV and ToBRFV viruses were used to identify the virus species in eggplant samples,and the phylogenetic analysis of the amplified sequences was carried out by using the maximum likelihood method.[Results]The specific band of 431 bp could be amplified only by the detection primers of ToCV.After sequencing,the sequence had the highest similarity with the ToCV sequence(KR072214),which was 99%,indicating that the virus in the eggplant sample was ToCV;Phylogenetic analysis showed that the ToCV eggplant isolates obtained in this study were clustered with ToCV isolates from Tianjin(KP893120),Shandong(KP072214),Spain(AAZ77648),Brasilia(AFN44466)and Greece(ABX89240),and afterwards,the phylogenetic relationship with ToCV isolates from Tianjin was analyzed.[Conclusion]It is confirmed for that tomato chlorosis virus is harmful to facility eggplants in Yingxia Township,Korla City,Xinjiang,and it was not co-infected with ToCV,TSWV,TMV and ToBRFV.关键词
