Optimization of processing technology and chemical components analysis of Zhujieshen(Panacis Japonici Rhizoma)based on HPLC and LC-Orbitrap-MS

Abstract
Objective To systematically identify the main chemical components of Zhujieshen(Panacis Japonici Rhizoma)and investigate the effects of moistening time,steaming time,and drying temperature on the content of the active ingredient ginsenoside Rg1 in its processed products[processed Zhujieshen(Panacis Japonici Rhizoma)],providing a scientific basis for quality control and processing technology optimization of Zhujieshen(Panacis Japonici Rhizoma).Methods Liquid chromatography-electrostatic field orbitrap high-resolution mass spectrometry(LC-Orbitrap-MS)technology,combined with the mzCloud and mzVault databases,was employed for the qualitative analysis of the main chemical components of Zhujieshen(Panacis Japonici Rhizoma).Processed Zhujieshen(Panacis Japonici Rhizoma)samples were prepared by setting different moistening times,steaming times,and drying temperatures.The samples were subjected to reflux extraction with diethyl ether.After recovering the diethyl ether,the samples were heated with methanol until the solution became colorless,followed by extraction with saturated n-butanol and preparation of the test sample solution with ammonia test solution.High-performance liquid chromatography(HPLC)was used to determine the content of ginsenoside Rg1 under different processing conditions,thereby optimizing the best processing technology.Results Under the processing conditions of 3 h of moistening,6 h of steaming,and a drying temperature of 90℃,the content of ginsenoside Rg1 in processed Zhujieshen(Panacis Japonici Rhizoma)reached the highest level.Through LC-Orbitrap-MS analysis,a total of 31 chemical components were identified,with structural types covering dammarane,ocotillol,and oleanane types,among others.Conclusion This study determined the optimal processing technology centered on 3 h of moistening,6 h of steaming,and drying at 90℃,which could significantly increase the content of ginsenoside Rg1 in processed Zhujieshen(Panacis Japonici Rhizoma).The established LC-Orbitrap-MS method is stable and reliable and can be used for the quality control and comprehensive development and utilization of Zhujieshen(Panacis Japonici Rhizoma).关键词
