核桃青皮水提物的抗氧化活性研究及机制探讨
Antioxidant activity of aqueous extract of green walnut husks and its mechanism of action

Abstract
Objective To investigate the in vitro antioxidant activity of aqueous extract of green walnut husks and its potential mechanism of action.Methods An aqueous extraction method was used to prepare the green walnut husk extract,and its in vitro antioxidant capacity was evaluated by 2,2-diphenyl-1-picrylhydrazyl(DPPH)radical scavenging assay,2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)cation radical scavenging assay,and ferric reducing antioxidant power(FRAP)assay;in combination with network pharmacology,key constituents and targets were screened,and molecular docking was performed to verify the binding capacity between constituents and targets.Results The in vitro antioxidant assays indicated that the extract exhibited marked concentration-dependent activity in all three evaluation systems,with an overall antioxidant capacity at an upper-middle level among similar studies.Network pharmacology analysis suggested that key components,including juglone,kaempferol,and quercetin,may act on multiple targets such as ESR2,MMP2,AKT1,and SRC,and were enriched in oxidative stress-related signaling pathways such as PI3K-Akt and HIF-1.Molecular docking showed that juglone had strong binding activity to ESR2(-7.3 kcal/mol),suggesting that it may synergize with flavonoid constituents to regulate redox homeostasis through the ESR2-PI3K-Akt signaling axis.Conclusion Green walnut husk extract has significant antioxidant activity,and its mechanism of action involves synergistic regulation across multiple constituents,multiple targets,and multiple pathways,providing a theoretical basis for the application of green walnut husks in the development of natural antioxidants and food-medicine homologous products.关键词
