垂穗披碱草近壳多胞叶斑病快速检测技术的建立与应用
Establishment and application of molecular detection for Parastagonospora nodorum-induced leaf spot disease in Elymus nutans

Abstract
To establish a rapid detection system for leaf spot disease of Elymus nutans caused by Parastagonospora nodorum,highly specific primers Pr-F/Pr-R targeting the conserved RPB2(RNA polymerase Ⅱ subunit 2)gene sequence of the pathogen was designed.An efficient,rapid,and stable PCR molecular detection technique was developed to identify Parastagonospora leaf spot in E.nutans.The results indicated:1)The detection system specifically amplified a 127 bp band from P.nodorum,and the sensitivity of Pr-F/Pr-R for detecting genomic DNA of P.nodorum was 100 pg/μL.2)The Pr-F/Pr-R primers can specifically detect P.nodorum in infected E.nutans leaf tissues,as well as in suspected diseased field-collected E.nutans plants.These results provide a technical basis for the early detection and targeted control of leaf spot disease in E.nutans.关键词
