Chemical and mechanical forces in TCR-peptide-MHC binding and their roles in viral infection

Abstract
Interaction between T cell receptor(TCR)and antigen peptide-major histocompatibility complex(pMHC)is a central component of adaptive cellular immune response.TCR-pMHC binding is governed not only by chemical forces,such as affinity,avidity and functional avidity(FA),but is also dynamically regulated by mechanical forces,which in turn influence binding conforma-tion and subsequent signal transduction.Therefore,process of T cell antigen recognition and activation are intricately linked to both chemical and mechanical forces.In context of viral infections,interplay of forces between TCR and pMHC exhibits distinct characteris-tics and plays differential roles in T cell responses,viral clearance and immune escape,depending on specific pathogen and infection state.This review focuses on elucidating mechanisms by which chemical and mechanical forces act in TCR-pMHC binding process and regulate T cell responses during viral infections.It further explores how mechanical forces can complement traditional chemical forces.By integrating insights from both chemical and mechanical perspectives,this review aims to provide novel ideas and strategies for immunotherapy of viral infectious diseases.关键词
