Identify the genes related to egg shell color of Jiuyuan black chickens based on RNA-seq technology

Abstract
In order to find out the genes related to the shell pigmentation of green shell eggs in the Jiuyuan black chickens,the experiment used transcriptome sequencing to study the oviduct tissues of green shell eggs and non-green shell eggs in the Jiuyuan black chickens,and screened differentially expressed genes.The results showed that 237 genes were significantly differentially expressed between the two groups(P<0.01),of which 123 genes were significantly up-regulated and 114 genes were significantly down-regulated.Gene Ontology annotation results showed that the differentially expressed genes were mainly enriched in transmembrane transport,substance transport,and transport activity.Kyoto Encyclopedia of Genes and Genomes enrichment analysis showed that eggshell pigment deposition and cell may signal with process,material transport pathway and so on.Four genes possibly related to pigmentation were screened out from the differentially expressed genes,which were SLC5A9,KCNF1,TYRP1 and CACNA1E.The results of fluorescence quantitative PCR showed that the expression trend of the four genes was consistent with the sequencing results,and there were extremely significant differences among different groups(P<0.01).The results showed that the four differentially expressed genes may play a role in the process of shell pigment deposition in the Jiuyuan black chicken,and this result provided a theoretical basis for the subsequent molecular selection of the Jiuyuan black chicken with green shell egg.关键词
