菜豆复合诱变突变体库构建及基因组分析
Construction of a Composite Mutagenized Snap Bean Mutant Library and Genomic Analysis

Abstract
Taking seeds of dwarf edible-podded snap bean(Phaseolus vulgaris)A18 as experimental material,a combined mutagenesis method involving nitrogen ion beam(4 × 1016 ions·cm-2)and 60Co-γ ray(80 Gy)irradiation was applied to investigate the effects of composite mutagenesis on phenotypic variation in snap bean and the genomic characteristics of the mutants,in order to create foundational materials for genetic breeding and functional genomics research in snap bean and provide reference for composite mutagenesis research in related crops.The results showed that a total of 374 mutant plants belonging to five phenotypic categories-leaf,flower/fertility,stem/plant architecture,seed,and growth period were screened in the M2 generation,with a mutation frequency of 6.34%.Nine stably inherited mutant lines were selected,advanced to the M3 generation,and subjected to whole-genome resequencing analysis.Significant differences were observed among mutants in nucleotide diversity(Pi:0.184-0.195)and genetic differentiation coefficient(Fst:0.111-0.172).The genetic similarity coefficients between mutants and the wild type ranged from 0.83 to 0.87.关键词
