Abstract

Taking Paeonia suffruticosa Andr.as the test materials,physicochemical properties,gene structure,conserved domains,chromosomal localization,secondary structures,promoter features,and expression patterns during leaf abscission were analyzed by bioinformatics analysis,in order to provide reference for molecular breeding of heat-tolerant varieties using the deciduous genes of peonies.The results showed that a total of 40 PsPME genes were identified,which were unevenly distributed across five chromosomes.Based on phylogenetic clustering,the PsPME genes were broadly categorized into three groups.The molecular weights of PsPME family members ranged from 12 015.85 to 65 720.35,with isoelectric points between 4.77 and 10.00,and aliphatic indices ranging from 56.07 to 102.19.Among them,24 were classified as alkaline proteins,and 6 were predicted to be unstable.The majority were identified as hydrophilic proteins.Subcellular localization analysis indicated that PsPME proteins may localize to various organelles including the cell wall,endoplasmic reticulum,chloroplast,cytoplasm,and nucleus.Conserved motif and domain analyses revealed that all 40 members contain the characteristic pectinesterase domain.Synteny analysis identified six pairs of orthologous genes between tree peony(Paeonia suffruticosa)and Arabidopsis thaliana.Signal peptides were predicted in 25%of the PsPME proteins.The promoter regions of PsPME family genes were found to contain numerous cis-acting elements associated with hormone response,low-temperature stress,anaerobic induction,and light responsiveness.Transcriptome analysis of the petiole abscission zone before and after its formation showed significant differential expression in 14 PsPME genes.Among these,PsPME18,PsPME21 and PsPME30 were significantly upregulated after the abscission zone formation,these three members may be involved in the regulation of tree peony defoliation.
