Abstract

Edwardsiella ictaluri is an important pathogen in aquaculture.In recent years,its increasing antibiotic resistance and cross-species transmission have raised serious concerns.Genomic studies of strains from different hosts are essential for exploring their pathogenic mechanisms,epidemiology,genetic diversity,and adaptive e-volution.To elucidate the antimicrobial resistance,genomic characteristics,and evolutionary relationships of an E.ictaluri strain YCH from yellow catfish,antimicrobial susceptibility testing and whole-genome sequencing and assembly were performed.Functional annotation was performed using multiple databases,and comparative and pan-genome analyses were conducted with nine other Edwardsiella strains.The results showed that the YCH ge-nome is 3.79 Mb in size,with a GC content of 57.42%,and comprises 3 439 coding sequences,25 rRNAs,and 99 tRNAs.A total of 34 genomic islands and 6 prophages were identified,while no CRISPR-Cas loci were detected.Antimicrobial susceptibility testing and resistance gene prediction indicated that the YCH strain might employ efflux pumps and target modification mechanisms to exhibit multidrug resistance to polymyxins,erythro-mycin,tetracyclines,and fluoroquinolones.Functional annotation revealed a large number of genes involved in metabolic pathways,with 3 564,3 078,and 3 384 genes assigned to GO,COG,and KEGG databases,respectively.Pan genomic analysis showed that YCH shares 2 294 conserved core genes with nine other Ed-wardsiella strains,while the number of strain-specific genes varied significantly(29~547).Phylogenetic analysis further revealed that YCH clusters closely with E.ictaluri Ei-VN5 in the same evolutionary branch.This study represents the first report of whole-genome sequencing and functional annotation of E.ictaluri strain YCH from yellow catfish,systematically revealing its genomic characteristics,functional distribution,antibiotic resistance,and evolutionary relationships.These findings provide an important insights into the pathogenicity and resistance mechanisms of E.ictaluri and offer valuable references for developing prevention and control strategies.
