基于Nrf2/HO-1信号通路探讨褪黑素对去卵巢大鼠氧化应激损伤的作用
The effect of melatonin on oxidative stress damage in ovariectomized rats based on the Nrf2/HO-1 signaling pathway

Abstract
Objective:To explore the mechanism of melatonin on oxidative stress damage in rats with rapid bone loss after ovariectomy based on the Nrf2/HO-1 signaling pathway.Methods:A total of 24 SD rats were randomly divided into a sham operation group,a model control group,and a melatonin-treated group,with 8 rats in each group.Bilateral oophorectomy was performed to replicate a rapid bone loss model,followed by gastric intervention for 12 weeks.Bone density was measured using a dual energy X-ray absorptiometry,and bone microstructure was observed using hematoxylin-eosin staining.ELISA was used to detect bone metabolism indicators including bone-specific alkaline phosphatase(BAP),tartrate-resistant acid phosphatase(TRAP),and oxidative stress indicators such as superoxide dismutase(SOD)and malondialdehyde(MDA).Real-time PCR and Western blot were used to detect the relative mRNA and protein expression levels of nuclear factor erythroid 2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)in the femur.Results:Compared with the sham operation group,the bone mineral density of rats in the model control group decreased significantly,indicating that the rat model of bone loss was successfully established.After 12 weeks of melatonin intervention,compared with the model control group,the BMD of the melatonin group was significantly increased(P<0.01);the number of trabeculae increased and the trabecular spacing shortened;the serum BAP and TRAP were both decreased(P<0.01);SOD was increased and MAD content was decreased(P<0.01);and the mRNA and protein expression levels of Nrf2 and HO-1 were significantly increased.Conclusion:Melatonin can activate the Nrf2/HO-1 signaling pathway,reduce oxidative stress damage,and improve bone loss caused by a sharp decrease in estrogen levels.关键词
