哮喘患儿miR-185-5p、CDC42表达水平与病情严重程度的关系
Relationship between miR-185-5p and CDC42 expression levels and disease severity in children with asthma

Abstract
Objective To investigate the expression levels of serum microRNA-185-5p(miR-185-5p)and cell division cycle protein 42(CDC42)in children with asthma and to explore their relationships with disease severity.Methods A total of 105 children with asthma hospitalized at the First Affiliated Hospital of Hebei North University from January 2022 to January 2024 were enrolled,along with 80 healthy children as controls.According to disease severity,asthmatic children were divided into a mild-to-moderate group(n=70)and a severe group(n=35).Serum levels of miR-185-5p and CDC42 were compared among the three groups.Pearson correlation analysis was used to evaluate the correlations between serum miR-185-5p and CDC42 levels and immunoglobulin E(IgE),interferon-γ(IFN-γ),and interleukin-4(IL-4).Logistic regression analysis was performed to identify factors influencing severe asthma.Receiver operator characteristic(ROC)curves were used to assess the diagnostic value of serum miR-185-5p and CDC42 levels for severe asthma.Results Serum miR-185-5p and CDC42 levels increased sequentially in the control group,mild-to-moderate group,and severe group,with statistically significant differences(F=241.335 and 95.161,respectively,P<0.05).Compared with the severe group,the mild-to-moderate group had significantly lower levels of IgE and IL-4 and higher levels of IFN-γ(t=4.179,4.585,and 5.205,respectively,P<0.05).Serum miR-185-5p and CDC42 levels were positively correlated with IgE and IL-4(r=0.490,0.479,0.541,and 0.361,respectively,P<0.05)and negatively correlated with IFN-γ(r=-0.358 and-0.410,respectively,P<0.05).A positive correlation was observed between miR-185-5p and CDC42 expression,and a potential targeting relationship was identified(r=0.512,P<0.001).Multivariate logistic regression analysis indicated that miR-185-5p and CDC42 were risk factors for severe asthma,whereas IFN-γ was a protective factor(OR=1.345,1.061,and 0.006,respectively,P<0.05).The area under the curve(AUC)values for serum miR-185-5p and CDC42 in diagnosing severe asthma were 0.793 and 0.885,respectively,and the combined AUC reached 0.898.Conclusion Serum miR-185-5p and CDC42 levels are significantly elevated in children with bronchial asthma and are closely associated with disease severity.Combined detection of miR-185-5p and CDC42 has high predictive value for assessing asthma severity in children.关键词
