Abstract

In order to establish a rapid and highly sensitive immunochromatographic method for the detec-tion of lumpy skin disease virus(LSDV)P32 protein antibodies,this study had developed immunochroma-tographic strips based on quantum dot(QDs)fluorescent microsphere labeling technology.The conjugation conditions of QDs with P32 protein and the optimal preparation parameters of the immunochromatographic strips were optimized.The specificity,sensitivity,reproducibility,and stability of the strips were evaluated,and the results were compared with those of a commercial detection kit.The results showed that this me-thod exhibited no cross-reaction with common viruses such as bovine viral diarrhea virus(BVDV),peste des petits ruminants virus(PPRV),bovine coronavirus(BCoV),and mycoplasma gallisepticum(MG),in-dicating good specificity.It could detect LSDV-positive serum diluted up to 1∶160,demonstrating high sensitivity.The strips maintained good detection performance after 120 days of storage,confirming excellent stability.Compared with the commercial detection kit,this method achieved a concordance rate of 86.00%and showed higher sensitivity,making it suitable for early screening of clinical samples.
