摘要

Abstract

To obtain microbial resources with efficient cellulose-degrading capacity,a strain of Bacillus velezen-sis was isolated and purified from the saline-alkali soil in Shihezi,Xinjiang,and designated as WL-04 in this study.The strain was identified through morphological observation,physiological and biochemical assays,and 16S rDNA sequence analysis.Single-factor experiments combined with response surface methodology were em-ployed to optimize fermentation conditions for enzyme production,while its straw degradation efficiency and stress tolerance were further evaluated.Phylogenetic analysis indicated that WL-04 belongs to Bacillus velezen-sis.The optimal enzyme production conditions were determined as temperature 39.5℃,time 25 h,and initial pH 6.1.Under these conditions,the endoglucanase activity reached 92.38 U/mL,representing a 95.60%increase compared to pre-optimization levels(P<0.01).Further evaluation of its degradation ability in a medium with cotton straw as the sole carbon source showed that after 15 days of cultivation,the overall weight loss rate of the substrate was 21.90%,with the degradation rate of the cellulose component reaching 26.20%,demonstrating good practical degradation potential.In terms of tolerance,WL-04 exhibited broad adaptability,capable of normal growth and enzyme production under pH 5.0-9.0,NaCl concentrations of 1.0%-6.0%,and a high tempera-ture of 50.0℃,indicating strong environmental adaptability.This study offers a promising strain with robust cel-lulose-degrading ability and strong environmental adaptability,facilitating the resource utilization of agricultural waste.
