1株蜜蜂源植物乳杆菌的分离鉴定及生物学特性分析
Isolation,Identification,and Characterization of a Bee-Derived Lactobacillus plantarum Strain

Abstract
In order to screen lactic acid bacteria strains inhibiting Ascosphaera apis as candidate probiotics for chalkbrood disease control,200 healthy honeybee guts were used as isolation source,and a lactic acid-bacterium(strain SZ107)was obtained through isolation,purification and screening on MRS(Man Rogosa Sharpe)medium.This strain was identified by morphological,biochemical and molecular biological meth-ods,and it was determined to be tolerannt to acid and bile salts,high sugar and high temperature,antioxi-dant capacity,inhibition rate against A.apis,antibiotic susceptibility,and safety for honeybee larvae were evaluated.The results showed that isolate SZ107 was identified as Lactobacillus plantarum,entered sta-tionary growth phase after 14 h,and the broth pH remained stable at approximately 3.8 after 20 h.Viable cell counts differed significantly under different pH(5.0,4.0,3.0,2.0)and bile salt concentrations(0.15%,0.30%,0.50%)(P<0.05),with survival rates of 107.73%,82.90%,62.12%,20.94%and 42.91%,10.95%,8.14%,respectively.Under 50℃treatment and different high sugar concentrations,the survival rates were all above 50%,and viable cell counts differed significantly among treatment groups(P<0.05).The ABTS radical scavenging rate reached 31.14%.At a concentration of 1×108 CFU/mL,the strain showed the highest inhibition rate against A.apis(45.91%).It exhibited resistance to norflox-acin,streptomycin and kanamycin,and susceptibility to five antibiotics including ampicillin,erythromycin,chloramphenicol,cefotaxime,amoxicillin.Feeding honeybee larvae with different concentrations showed no significant difference in survival rate compared with the control group(P>0.05).In conclusion,isolate SZ107 exhibits favorable growth characteristics,tolerance,antioxidant capacity and antifungal activity a-gainst A.apis,is safe and non-toxic to honeybee larvae,and has potential as a probiotic for chalkbrood dis-ease prevention and control.关键词
