Detection and Genetic Sequence Analysis of Porcine Epidemic Diarrhea Virus on Selected Large-scale Swine Farms in Sichuan Province

Abstract
In order to investigate the epidemiological characteristics and genetic variation of porcine epidemic diarrhea virus(PEDV)in Sichuan Province,a total of 123 fecal samples were collected from pigs with diarrhea on 18 large-scale farms across 12 regions in the province from 2024 to 2025,to detect PEDV nucleic acid using a fluorescence quantitative RT-PCR assay.105 positive samples were detected,with a sample positivity rate of 85.37%and a farm positivity rate of 100%;8 complete PEDV genomes were successfully obtained via high-throughput sequencing,each originating from a different region.It was found that,by phylogenetic analysis on complete genome and spike(S)gene sequences,the 8 strains belonged to G2c(n=5),G2b(n=2)and S-INDEL(n=1)subtypes,respectively;and multiple amino acid mutations were identified in the translated S protein sequences of all the strains.In conclusion,PEDV was the primary pathogen responsible for porcine diarrhea on large-scale farms in Sichuan Province,and multiple genotypes were co-circulating,but dominated by G2c subtype;and the efficacy of current vaccines might be reduced by numerous mutations in key antigenic sites of the S protein.Therefore,PEDV should be increasingly monitored,and novel broad-spectrum vaccines including those against G2c strain should be developed as soon as possible,to curb the ongoing spread of PEDV.关键词
