摘要

Abstract

In order to establish an efficient in vitro rapid propagation system for the Corymbia citriodo-ra subsp.variegata,stem segments of sterile seedlings of this species were used as explants.The key steps such as non-stem bud induction,proliferation,rooting medium,and acclimatization and trans-plantation were systematically studied.The results showed that the disinfection of Corymbia citriodo-ra subsp.variegata seeds using a combination of 75%alcohol and 0.1%mercuric chloride resulted in high seed survival rate and low contamination rate.The optimal medium for non-stem bud induction was:2 DCR+1.00 mg/L 6-BA+0.01 mg/L NAA,with an induction rate of 100%,without brown-ing or glassification phenomena.The optimal medium for subculture proliferation was:2 DCR+0.30 mg/L 6-BA+0.05 mg/L NAA,with a proliferation coefficient of 15.14,and the seedlings grew robustly without the need for vigorous seedling cultivation.The optimal rooting medium was:1/2 DCR+1.00 mg/L IBA+0.50 mg/L NAA+0.50 mg/L ABT,with a rooting rate of 93.33%and an average root length of 2.96 cm.The most suitable substrate ratio for transplantation was:Vhumussoil∶Vred soil∶Vpeat soil=1∶1∶1),with an survival rate of 95.24%.
