Abstract

This article focused on investigating the in vitro inhibitory effects of honokiol on Cutibacterium acnes(C.acnes)and its mechanism of action.The antibacterial activity was assessed using the agar diffusion method,broth microdilution method,and time-kill curve.The effects of honokiol on C.acnes surface hydrophobicity,biofilm formation,and metabolic activity were analyzed through the microbial adhesion to hydrocarbons(MATH)test,crystal violet staining method,and MTT assay.Additionally,morphological changes in bacterial cells were observed using scanning electron microscopy(SEM).Results demonstrated that honokiol exhibited significant concentration-dependent antibacterial effects against C.acnes,with a minimum inhibitory concentration(MIC)of 15.63 μg/mL and a minimum bactericidal concentration(MBC)of 31.25 μg/mL.At MIC or higher concentrations,complete bacterial growth inhibition was achieved within 56 hours.Multidimensional experiments revealed that honokiol likely exerts its antibacterial effects by altering bacterial surface hydrophobicity,inhibiting biofilm formation,and disrupting cell integrity.The study indicated that honokiol displays multi-target antibacterial effects against C.acnes,providing reference for the development of natural antibacterial agents in acne treatment.
