Study on the effects and mechanisms of irreversible electroporation on ovarian cancer

Abstract
Objective To explore the cytotoxic efficacy and underlying mechanisms of irreversible electroporation(IRE)on ovarian cancer cells in vitro and in vivo models.Methods SK-OV-3 ovarian cancer cells were treated with IRE under different electric field strengths,pulse widths,and pulse numbers,with untreated cells serving as the control.Cell viability,apoptosis,and necrosis were evaluated using the CCK-8 and flow cytometry.To investigate the mechanisms of IRE-induced apoptosis and necrosis in SK-OV-3 cells,the expression of apoptosis-associated proteins was examined using Western blot and qRT-PCR.Additionally,intracellular ROS levels,mitochondrial membrane potential,ATP release and Ca2+concentration were assessed.By constructing a subcutaneous tumor model using SK-OV-3 cells and applying different electric field intensities(0,400,800 and 1 000 V/cm)to treat the tumors—with the 0 V/cm group serving as the control.A subcutaneous SK-OV-3 xenograft model was established to evaluate the inhibitory effect of IRE on tumor growth and its impact on anti-tumor immune response in vivo.Tumor volume was monitored for 24 d post-IRE treatment,and serum concentrations of IL-1 β,TNF-α,and IFN-γwere measured to preliminarily assess the systemic immunodulatory effects.Results The cytotoxic effect on SK-OV-3 cells was enhanced with increasing electric field strength and pulse number,which was accompanied by a rise in the proportion of necrotic cells.When the electric field intensity reached 800 V/cm,almost no viable cells were detected[viable cell proportion:(7.14±0.16)%].The subcutaneous tumor model in animals showed that,compared with the control group,IRE treatment significantly inhibits the growth of subcutaneous tumors in mice.Under conditions of 800 V/cm field strength and 100 μs pulse width,50 pulses achieve the optimal killing effect on SK-OV-3 cells.Conclusions IRE can induce SK-OV-3 cell death by perturbing intracellular homeostasis.The proportion of necrotic cells increases with higher electric field strength and pulse number.关键词
