滩羊不同时期皮肤组织KRTAP1-1、KRTAP6-1和TCHH基因表达模式研究
Study on the Expression Pattern of KRTAP1-1,KRTAP6-1 and TCHH Genes in Skin Tissue of Tan Sheep Lambs at Different Periods

Abstract
To explore the expression pattern of KRTAP1-1,KRTAP6-1 and TCHH genes in the skin tissues of Tan sheep lambs at different periods,quantitative real-time PCR(qRT-PCR)and Western blot(WB)were employed to determine the expression level of KRTAP1-1,KRTAP6-1 and TCHH mRNA and their encoded proteins in the skin of Tan sheep at day 0(control group),35 and 60 of age.The results showed that the expression level of KRTAP1-1 gene was gradually increased with the growth of age,and its expression level at day 35 and 60 was extremely significantly higher than that at day 0(P<0.01),and the expression level at day 60 was significantly higher than that at day 35(P<0.05).With the growth of age,the expression level of KRTAP6-1 gene was also gradually increased,and its expression level at day 60 was extremely significantly higher than that at day 0 and 35(P<0.01),but there was no significant difference at day 0 and 35(P>0.05).Contrast to the other two genes,the expression level of TCHH gene was gradually decreased with the growth of age,and its expression level at day 0 was significantly higher than that at day 35(P<0.05),extremely signifi-cantly higher than that at day 60(P<0.01),and the expression level at day 35 was significantly higher than that at day 60(P<0.01).The WB results showed that the expression level of KAP1-1 protein in the skin tissue of Tan sheep at day 35 was significantly higher than that at day 0(P<0.05),and extremely significantly higher than that at day 60(P<0.01).With the growth of age,the expression level of TCHH protein was gradually decreased,and its protein expression level at day 0 was significantly higher than that at day 60(P<0.01),the expression level at day 35 was significantly higher than that at day 60(P<0.05),but there was no significant difference at day 0 and 35(P>0.05).关键词
