烟草NtJAZ11参与调控表皮毛发育的影响研究
Effects of NtJAZ11 on Tobacco Trichomes Development

Abstract
To explore the function of NtJAZ11 on the development of trichomes development in tobacco,based on homologous cloning,sequence characteristics and expression pattern of NtJAZ11,glandular trichome phenotype and its related genes expression patterns in both NtJAZ11 gene knockout and overexpression transgenic lines were analyzed.The results showed that 2 homologous genes of NtJAZ11,NtJAZ11-1 and NtJAZ11-2,were cloned in tobacco,whose coding sequence lengths were 1 143 and 1 182 bp,respectively,and both containing the typical TIFY and Jas domains.The subcellular localization showed that NtJAZ11 fusion protein was localized in the nucleus and cell membrane.Expression analysis clarified that NtJAZ11 had the highest expression level in leaves and was strongly induced by methyl jasmonate treatment.The density of long and short glandular trichomes in NtJAZ11 overexpression lines decreased significantly,and the density of nonglandular trichomes increased obviously.While the density of long and short glandular trichomes in NtJAZ11 gene knockout lines increased significantly,and the density of nonglandular trichomes remained unchanged.The qRT-PCR analysis verified that the relative expression levels of glandular trichomes development related gene including NtWo,NtCycB2,NtMIXTA,NtMYC2,NtHD8,NtHD9 and NtHD12 were all significantly affected by NtJAZ11.Above results indicated that NtJAZ11 played a negative role in the density of long and short glandular trichomes,and the positive one in the formation of nonglandular trichomes.关键词
