干扰素诱导跨膜蛋白1促进猪丁型冠状病毒复制作用研究
Interferon-induced transmembrane protein 1 facilitates porcine deltacoronavirus replication

Abstract
This study aimed to investigate the role of interferon-induced transmembrane protein 1(IFITM1)in the replication of porcine deltacoronavirus(PDCoV).An IPEC-J2 cell model infected with PDCoV was established,and the transcriptional and protein expression levels of IFITM1 were detected by qPCR and Western-blot,respectively.The effect of IFITM1 on viral replication was observed after siRNA-mediated IFITM1 silencing,and the specific role of IFITM1 in the viral replication cycle was further analyzed.Key viral proteins regulating IFITM1 expression were screened by transfection with plasmids expressing different PDCoV structural proteins,and the subcellular localization of IFITM1 was analyzed using im-munofluorescence.The results showed that PDCoV infection significantly upregulated IFITM1 expression in IPEC-J2 cells.Silencing IFITM1 inhibited PDCoV replication,and IFITM1 was found to primarily promote viral internalization and early replication.Further studies revealed that the PDCoV S protein was the key factor inducing IFITM1 upregulation,and immunofluorescence assay demonstrated their colocalization in the cytoplasm and cytomembrane.In conclusion,IFITM1 significantly promotes PDCoV replication,providing new theoretical insights into the study on pathogenic mechanism of PDCoV.关键词
