Study on biofilm activity of novel selenomaleimide compound CY-162-13 against Staphylococcus aureus

Abstract
Objective To study the effect of the novel selenomaleimide compound CY-162-13 at sub-inhibitory concentrations on the biofilm-forming ability of Staphylococcus aureus,providing a theoretical basis for the development of antimicrobial drugs.Methods Staphylococcus aureus strains preserved in the First Affiliated Hospital of Wenzhou Medical University laboratory for a long time were revived.The minimum inhibitory concentration(MIC)of CY-162-13 was determined using the broth microbroth dilution method.The impact of CY-162-13 at sub-inhibitory concentrations on the biofilm formation ability of Staphylococcus aureus was assessed using the crystal violet staining and confocal laser scanning microscopy.The adhesion quantity of Staphylococcus aureus and the generation level of polysaccharide intercellular adhesin(PIA)under the action of CY-162-13 were detected by cell adhesion assay and enzyme-linked immunospot assay,respectively.Results The CY-162-13 at a concentration of 1.0 μg/mL inhibited the biofilm formation of methicillin-resistant Staphylococcus aureus,decreased initial attachment and repressed the production of PIA.Conclusion The compound CY-162-13 at a sub-inhibitory concentration inhibits the biofilm formation by decreasing Staphylococcus aureus adhesion and PIA production.This provides a new approach for the clinical treatment of infections caused by Staphylococcus aureus biofilms,and is worth further development.关键词
