Optimization and Establishment of Regeneration System of Cucurbita spp.

Abstract
Taking different genotypes of Cucurbita spp.as the experimental materials,tissue culture techniques was used to investigate the effects of explant types,genotypes,and varying hormone combinations on the induction,elongation,and rooting of adventitious buds,in order to establish a stable and highly efficient regeneration system for Cucurbita spp.,thereby providing a reference for genetic transformation studies in this species.The results showed that there were significant differences in the induction rates of adventitious buds from different types of explants.The induction rate of adventitious buds from 1/2 leaf nodes was significantly higher than that from 1/4 leaf nodes.The regeneration efficiency with different genotypes was significantly different.C.pepo inbred line'19',C.moschata inbred line'C.moschata No.2'and C.maxima inbred line'282'with high regeneration efficiency were selected to carry on the subsequent optimal hormone ratio test.The combination of MS+6-BA 2.0 mg·L-1+ABA 0.5 mg·L-1 was the optimal medium for bud induction with a 88.3%induction rate of'19'.The combination of MS+6-BA 2.0 mg·L-1+ABA 1.0 mg·L-1 was the optimal medium for bud induction with a 85.0%induction rate of'C.moschata No.2'.The combination of MS+6-BA 1.0 mg·L-1+ABA 1.0 mg·L-1 was the optimal medium for bud induction with a 100.0%induction rate of'282'.MS+KT 0.2 mg·L-1 was the optimal medium for adventitious bud elongation with strong shoots and growth in the three genotypes Cucurbita spp.MS without added hormones was the optimal medium for shoot rooting with the long and developed roots.关键词
