寿胎丸对人早孕滋养细胞增殖与凋亡的影响及相关机制研究
Effect of Shoutai Pill on Proliferation and Apoptosis of Human Trophoblast Cells in Early Pregnancy and Its Related Mechanism

Abstract
Objective:To investigate the effects of the serum containing Shoutai pill on the proliferation,PCNA expression,migration and apoptosis of human trophoblasts in early pregnancy,and the secretion of the chemokines CXCL12 and CXCL16.Methods:The human trophoblasts of early pregnancy were isolated and cultured in vitro,and were intervened with low,medium and high doses of serum containing Shoutai Pill,with negative serum group(the control group)and desogestrel group as positive control groups respectively.CCK-8,flow cytometry,Transwell,AnnexinV-FITC/PI double labeled flow cytometry and ELISA were used to determine the effects of serum containing drugs on the proliferation activity,PCNA expression,migration ability,total apoptosis,chemokines CXCL12 and CXCL16 of human trophoblasts in early pregnancy.Results:Compared with the negative group,after the intervention of the serum containing Shoutai Pill,the proliferation activity,invasion and migration ability of human trophoblasts in early pregnancy were significantly enhanced,the apoptosis rate was significantly decreased,and the contents of chemokines CXCL12 and CXCL16 were significantly increased(P<0.05).Compared with the low-dose group of Shoutai Pill,the proliferation activity,invasion and migration ability of trophoblast cells in the middle and high-dose groups of Shoutai Pill were significantly enhanced,and the contents of chemokines CXCL12 and CXCL16 were significantly increased(P<0.05).Conclusion:The medicated serum of Shoutai Pill can improve the proliferation activity,invasion and migration ability of human trophoblasts in early pregnancy,and the content of chemokines CXCL12 and CXCL16,reduce the apoptosis rate and promote cell proliferation.关键词
