Effects of electroacupuncture on blood pressure circadian rhythm,myocardial inflammatory response and BMAL1/PPARα/NF-κB pathway in spontaneously hypertensive rats

Abstract
Objective:To observe the effects of electroacupuncture on blood pressure circadian rhythm,myocardial inflamma-tory response,and BMAL1/PPARα/NF-κB pathway in spontaneously hypertensive rats(SHR),and to explore the possible mechanisms of electroacupuncture to improve blood pressure circadian rhythm and cardiac injury in SHRs.Methods:A total of 15 spontaneously hypertensive rats were randomly divided into the model group,the electroacupuncture group and the Western medi-cine group,with 5 rats in each group.Another 5 Wistar Kyoto(WKY)rats were used as the control group.In the electroacupunc-ture group,electroacupuncture was applied to"Baihui",bilaterally"Neiguan"and bilaterally"Zusanli"."Neiguan"and"Baihui"were connected with electroacupuncture instrument for 20 min each time,and the treatment was continuoued for 6 weeks.The Western medicine group was given 7.2 mg/kg valsartan(Daiwen)by gavage for 6 weeks.Before and after the intervention,blood pressure circadian rhythm changes were measured in each group.H&E and Masson staining were used to observe the morphology and pathological changes of rat heart tissue.Immunohistochemistry was used to observe the expression of BMAL1 and PPARα in the heart of rats in each group.Quantitative real-time PCR was used to detect the transcription levels of BMAL1,PPARα,and IL-1β in the heart of rats.Western blot was used to detect the protein levels of BMAL1 and PPARα in the heart of rats.ELISA was used to detect IL-1β content in the heart of rats.Results:Compared to the control group,the model group had a weakened cir-cadian rhythm of blood pressure,more inflammatory cells infiltration in the heart,an increased level of cardiac fibrosis(P<0.05),a decreased level of BMAL1 and PPARα transcription and protein expression(P<0.05),and an increased level of p-NF-κB and IL-1β expression(P<0.05).Compared to the model group,in the electroacupuncture group,the circadian rhythm of blood pres-sure was restored,the degree of myocardial fibrosis was reduced(P<0.05),the expression levels of BMAL1 and PPARα were elevated(P<0.05),and the expression levels of p-NF-κB and IL-1β were reduced(P<0.05);In the Western medicine group,the circadian rhythm of blood pressure did not show significant restoration,the degree of cardiac fibrosis was reduced(P<0.05),and the BMAL1 transcription level and positive expression were elevated(P<0.05),PPARα protein expression was elevated(P<0.05),and IL-1β transcription level as well as p-NF-κB and IL-1β protein expression levels were reduced(P<0.05).Conclu-sion:Electroacupuncture can regulate the circadian rhythm of blood pressure and improve cardiac inflammatory response in hyper-tensive rats,and the mechanism may be related to the activation of BMAL1-mediated PPARα inhibition of the NF-κB signaling pathway to reduce the secretion of IL-1β by cardiomyocytes.关键词
