犹素化修饰在肿瘤发生发展中作用的研究进展
Research advance on UFMylation in tumorigenesis and progression
摘要
Abstract
UFMylation is a novel ubiquitin-like modification.In this process,the mature ubiquitin-fold modifier 1(UFM1)molecule is covalently conjugated to substrate proteins via a cascade catalyzed by the specific activating enzyme E1(UBA5),conjugating enzyme E2(UFC1),and ligase E3(UFL1),thereby regulating the functions of target proteins.Research indicates that UFMylation plays significant roles in various biological processes,including the regulation of protein stability,endoplasmic reticulum stress,and DNA damage repair.Abnormal regulation of UFMylation is closely associated with the development of various human tumors.On one hand,UFMylation can accelerate tumor progression by activating oncogene signaling pathways,on the other hand,it can exert tumor-suppressive effects by maintaining genomics stability and inhibiting oncogene activity,suggesting a dual role of UFMylation in tumor regulation.This paper reviews the mechanism,biological functions of UFMylation,and its role in tumor development,while also discussing the prospects of targeting UFMylation for cancer therapy.关键词
