The expression of MX1 gene in pancreatic cancer tissues and its impact on their biological behavior

Abstract
Objective To investigate the expression of myxovirus resistance 1(MX1)protein in pancreatic can-cer tissues and its relationship with clinicopathological features of pancreatic cancer patients,as well as its effects on the proliferation,migration,and invasion of pancreatic cancer cells.Methods Utilize the GEPIA online database to predict the differential expression of MX1 between pancreatic cancer tissues and adjacent non-cancerous tissues,as well as its correlation with prognosis.Employ immunohistochemistry,RT-qPCR,and Western blotting to compare MX1 expression levels in adjacent non-cancerous tissues versus pancreatic cancer tissues and cell lines.Use the χ² test to analyze the association between MX1 expression in pancreatic cancer tissues and clinicopathological features.Apply CCK-8,colony formation,EdU,and Transwell assays to assess the effects of MX1 knockdown on the prolifera-tion,invasion,and migration of PDAC cells.Perform GSEA enrichment analysis on MX1 and use Western blotting to evaluate the impact of MX1 on proteins related to the EMT pathway.Results Data from the GEPIA database and im-munohistochemistry showed that MX1 was highly expressed in PDAC tissues compared with adjacent non-tumor pan-creatic tissues(P<0.05).The expression of MX1 was associated with lymph node metastasis and vascular invasion in pancreatic cancer,which could serve as an independent prognostic risk factor for pancreatic cancer.Compared with normal human pancreatic ductal epithelial cells,pancreatic cancer cell lines exhibited higher expression of MX1.Knockdown of MX1 reduced the proliferation,invasion,and migration capabilities of PDAC cells.GSEA enrichment analysis indicated that MX1 might exert its effects in pancreatic cancer through multiple pathways.After MX1 knock-down,the expression of the epithelial marker E-cadherin was upregulated,while the expressions of the mesenchymal markers N-cadherin and vimentin were downregulated.Conclusion MX1 is highly expressed in pancreatic cancer tis-sues,and its elevated expression is associated with poor patient prognosis.It may promote the proliferation and migra-tion of pancreatic cancer cells by regulating the EMT pathway,suggesting its potential as a diagnostic and prognostic biomarker target in pancreatic cancer.关键词
