Role and molecular mechanisms of CMTM4 in colorectal carcinogenesis

Abstract
Objective To investigate the role of chemokine-like factor family member 4(CMTM4)in the devel-opment of colorectal cancer(CRC)and to explore its underlying molecular mechanisms.Methods The expression of CMTM4 in CRC tissues and normal colorectal tissues was examined using immunohistochemistry and reverse transcrip-tion polymerase chain reaction(RT-PCR).The associations between CMTM4 expression and clinicopathological pa-rameters as well as patient prognosis were analyzed.The effects of CMTM4 on CRC cell proliferation,migration,and invasion were evaluated CCK-8 proliferation assays and Transwell migration and invasion assays.In addition,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses were performed on CMTM4 co-expressed genes,and correlations between CMTM4 expression and immune cell infiltration were assessed.Results Immunohistochemistry and RT-PCR analyses demonstrated that CMTM4 expression was significantly lower in CRC tis-sues than in normal tissues.Low CMTM4 expression was significantly associated with advanced T stage,Duke stage,and poor prognosis in patients with CRC.CCK-8 assays showed that CMTM4 overexpression significantly inhibited CRC cell proliferation.Transwell assays revealed that CMTM4 overexpression markedly reduced the migratory and in-vasive abilities of CRC cells.GO and KEGG analyses indicated that CMTM4 was mainly involved in immune-related and metabolism-related pathways.Furthermore,CMTM4 expression was positively correlated with immune cell infiltra-tion levels and with the expression of multiple immune cell marker gene sets.Conclusion CMTM4 is involved in the development and progression of colorectal cancer and may serve as a potential molecular therapeutic target for CRC.关键词
