The mechanism of silencing P4HA1 to influence the proliferation and invasion of HT-29 and SW620 colorectal cancer cells

Abstract
Aim To explore the mechanism of silencing P4HA1 on the proliferation and invasion of colorectal cancer(CRC)cells HT-29 and SW620.Methods The expression level of P4HA1 in CRC tissues and its relationship with patient prognosis were analyzed through the GEPIA and Kaplan-Meier Plotter databases.Immunohistochemistry was used to detect the expression of P4HA1 in cancerous and adjacent normal tissues from 90 CRC patients.The proliferation,invasion,and migration abilities of HT-29 and SW620 cells after P4HA1 silencing were assessed using cell proliferation experiments,plate cloning experiments,and Transwell experi-ments.Western blotting was employed to detect the expression levels of epithelial-mesenchymal transition(EMT)-related proteins[E-cadherin,N-cadherin,and vimentin]in SW620 cells after P4HA1 silencing.Results The expression level of P4HA1 in CRC tissues was higher than that in normal tissues adjacent to the cancerous tissues.High expression of P4HA1 was a poor prognostic factor for overall survival in CRC patients(P<0.05).After silencing P4HA1,the proliferation,invasion,and migration abilities of CRC HT-29 and SW620 cells were significantly decreased(P<0.05).After silencing P4HA1,the expression of N-cadherin and vi-mentin in CRC SW620 cells were downregulated(P<0.05),while the expression of E-cadherin was upregulated(P<0.05).Conclusion Silencing P4HA1 inhibits the proliferation and invasion abilities of CRC HT-29 and SW620 cells,and its mechanism may be related to regulating the EMT process.关键词
