摘要

Abstract

[Objectives]This paper aimed to investigate the physiological effects of N-difluoromethyl imidazole selenourea(FASU-41)on inhibiting the growth and development of diamondback moth larvae(DBM).[Methods]The test insects were treated by the leaf dipping method.The contents of energetic substances including glucose,alginate,and triglycerides were determined by kit assay;the differentially expressed genes(DEG)were analyzed by transcriptome sequencing,and the results were verified by qPCR.[Results]FASU-41 resulted in symptoms such as sluggish larval activity,molting disorders,severe damage to midgut tissues,developmental malformations,and a significant decrease in the levels of energy substances such as glucose,alginate,and triglycerides in the larvae.GO annotation and KEGG enrichment analysis showed that the DEG were significantly enriched in pathways associated with redox reactions,carbohydrate metabolism and metabolism of epidermal structural substances.Transcriptome sequencing results showed that FASU-41 inhibited the expression of several antioxidant enzyme genes in the peroxisomes of the diamondback moth,leading to a decrease in its antioxidant capacity;down regulation of gene expression of enzymes involved in starch and sucrose hydrolysis weakened the digestive ability of the test insects toward starch and sucrose;it inhibited glycogen breakdown and glycolysis processes,leading to a decrease in glucose and trehalose content,and hindering energy conversion and supply;it inhibited the entire process of branched chain amino acid and fatty acid breakdown metabolism,further blocking the body's energy source;it also inhibited the hydrolysis of juvenile hormones and the synthesis of molting hormones in the test insects,resulting in molting disorders,delayed growth and development,and abnormal metamorphosis.[Conclusions]N-difluoromethyl imidazole selenourea is a kind of potential insecticidal lead compound targeting the pathway of growth and development.
