Predictive Value of Peripheral Blood IL-8 and IL-17 Levels Detected by Flow Cytometry for Preoperative Lymph Node Metastasis of Triple-Negative Breast Cancer

Abstract
Objective To explore the predictive value of peripheral blood interleukin-8(IL-8)and interleukin-17(IL-17)levels for preoperative lymph node metastasis in patients with triple-negative breast cancer(TNBC).Methods 110 TNBC patients admitted to the breast surgery department of our hospital from September 2023 to January 2025 were selected as study subjects.They were divided into the lymph node metastasis group(42 cases)and the non-lymph node metastasis group(68 cases)based on postoperative pathological results.Preoperative peripheral blood IL-8 and IL-17 levels were detected using flow cytometry in both groups.Clinical pathological data were collected and subjected to univariate and multivariate Logistic regression analysis.Receiver operating characteristic(ROC)curves were plotted to evaluate the predictive performance of IL-8,IL-17,and their combined detection.Results The levels of IL-8 and IL-17 in peripheral blood of the lymph node metastasis group were significantly higher than those of the non-lymph node metastasis group(P<0.001).Univariate analysis showed that tumor diameter,histological grade,carcinoembryonic antigen(CEA),and carbohydrate antigen 15-3(CA15-3)were associated with preoperative lymph node metastasis in TNBC(P<0.05).Multivariate Logistic regression analysis indicated that the levels of IL-8 and IL-17 in peripheral blood were independent risk factors for preoperative lymph node metastasis of TNBC(P<0.001).ROC curve analysis revealed that the area under the curve(AUC)for individual detection of IL-8 and IL-17 was 0.713 and 0.744,respectively,while the AUC for combined detection was 0.857;the combined predictive efficacy was higher.Conclusions Elevated levels of IL-8 and IL-17 in peripheral blood are closely associated with preoperative lymph node metastasis of TNBC.Combined detection of these two markers can improve the accuracy of predicting lymph node metastasis,providing a basis for clinical surgical planning and prognosis evaluation.关键词
