Mechanism of NLRP3 inflammasome induced-pyroptosis in Alzheimer disease

Abstract
Alzheimer disease(AD)is a common neurodegenerative disorder characterized by cognitive decline and progressive memory loss.Pyroptosis releases inflammatory factors such as interleukin-1β and interleukin-18,which then exacerbate inflammation and neuronal death,and intensify pathological changes such as amyloid β-protein deposition and excessive phosphorylation of Tau protein in AD.The NLRP3 inflammasome is an important inducer of pyroptosis.Studies have shown that amyloidβ-protein deposition,excessive phosphoryla-tion of Tau protein,mitochondrial dysfunction,oxidative stress,endoplasmic reticulum stress,and lysosomal damage can all activate the NLRP3 inflammasome.This article reviews the pathological connection between NLRP3 inflammasome-induced pyroptosis and AD,and summarizes the amelioration of the pathological progression of AD to provide new therapeutic approaches for AD in the future.关键词
