Effect of enzymatic hydrolysis on the structure and bioactivity of hawk tea pol-ysaccharides

Abstract
Objective This study examined the impact of enzymatic hydrolysis on the structural characteristics and bioactivities of polysaccharides from hawk tea.Methods The polysaccharide content,protein content,and uronic acid content were determined by the sulfuric acid-phenol method,BCA assay,and m-hydroxydiphenyl method,respectively.The monosaccharide composition was analyzed by PMP derivatization followed by high-per-formance liquid chromatography(HPLC),the molecular weight was determined by high-performance gel perme-ation chromatography(HPGPC),and the functional group structure was characterized by fourier transform infra-red spectroscopy(FT-IR).The inhibitory effect of polysaccharides on the proliferation of colon cancer cells was evaluated using the MTT assay,while the in vitro anti-inflammatory activity was screened by quantitative poly-merase chain reaction(qPCR).Antioxidant capacity was assessed through DPPH and ABTS free radical scaven-ging assays,and the hypoglycemic potential was examined using an α-glucosidase inhibition model.Results En-zymatic hydrolysis did not alter the types of monosaccharides in hawk tea polysaccharides but changed the molar ratios of the monosaccharide components,with glucose and arabinose contents showing the most significant varia-tions.The polysaccharide fraction treated with papain exhibited the strongest inhibitory activity against colon cancer cell proliferation and the highest α-glucosidase inhibitory capacity.Conclusion The study demonstrates that all five enzymatic methods effectively alter the structural properties of polysaccharides from hawk tea.Poly-saccharides treated with papain showed improved anti-colon cancer activity and hypoglycemic effects,providing fundamental data to support further research on their targeted enzymatic modification.关键词
