艾灸"足三里"对慢性不可预知温和应激致抑郁大鼠血清和结肠中5-羟色胺含量及肠道菌群的影响
Effect of Moxibustion at Zusanli on the Content of 5-hydroxytryptamine in Serum and Colon and Intestina Microbiota in a Rat Model of Depression Induced by Chronic Unpredictable Mild Stress

Abstract
Objective To investigate the effect of moxibustion at"Zusanli"on the content of 5-hydroxytryptamine(5-HT)in serum and colon and intestinal microbiota in a rat model of depression induced by chronic unpredictable mild stress(CUMS).Methods A total of 24 specific pathogen-free male Sprague-Dawley rats were randomly divided into blank group,model group,drug group,and moxibustion group,with 6 rats in each group.After the model of CUMS-induced depression was successfully established,the rats in the drug group and the moxibustion group were given fluoxetine and moxibustion,respectively,for 7 days.The sucrose preference test,the open field test,and the forced swimming test were used to assess behavioral changes;ELISA was used to measure the content of 5-HT in the colon and serum of rats;16S rRNA high-throughput sequencing was used to analyze the abundance of intestinal microbiota.Results Moxibustion at"Zusanli"significantly improved the degree of sucrose preference in rats with CUMS-induced depression(P＜0.05),increased the total distance traveled in the open field test(P＞0.05),significantly shortened immobility time in the forced swimming test(P＜0.05),and significantly increased the content of 5-HT in blood and colon(P＜0.05).In addition,moxibustion intervention could improve the ACE index and Chao 1 index of intestinal microbiota(P＞0.05),increase the abundance of Bacteroidetes,Spirochaetes,Campylobacter,Proteobacteria,and Actinobacteria,and re-duce the abundance of Desulfobacterota and Firmicutes.Conclusion Moxibustion at"Zusanli"can regulate the level of 5-HT in the blood and colon of rats with CUMS-induced depression,promote the production of probiotics,and inhibit the proliferation of opportun-istic pathogens,thereby maintaining intestinal homeostasis and improving depression-like behavior.关键词
