Abstract

[Objective]The role of Lhc genes in Nicotiana benthamiana under the adversity stress was explored to provide gene resources and theoretical support for breeding N.benthamiana germplasm resources with the high photosynthetic efficiency and strong stress resistance.[Method]The physicochemical properties,subcellular localization,gene structure,conserved motifs,promoter cis-regulatory elements,phylogenetic relationships and stress response pattern of the identified members of Lhc gene family in N.benthamiana at a whole-genome level were analyzed.[Result]51 Nblhcs genes identified from N.benthamiana were divided into 5 sub-families of CP24,CP26,CP29,Lhca and Lhcb.The amino acid number,relative molecular mass and isoelectric point of 47 genes located in the chloroplast were 175-986,19.31-105.18 kDa and 4.85-9.22 respectively.The promoters of Nblhcs genes in N.benthamiana contained numerous light-responsive elements,hormone-regulated elements of abscisic acid,jasmonic acid and salicylic acid,and stress-responsive elements against low temperature and drought.Based on promoter cis-regulatory elements,15 genes were selected for RT-qPCR verification under stress response.Under the stresses of darkness,low temperature,drought,jasmonic acid,and potato PVY virus,the expression level of 13 genes changed significantly,indicating that Nblhcs genes in N.benthamiana were involved in stress response.Under PEG treatment,the relative expression level of Nblhc28 was upregulated by 31 times,suggesting that it was a key gene of drought resistance in N.benthamiana.[Conclusion]The sub-family members of Nblhcs in N.benthamiana have the similarity in genetic structure,and their promoters are rich in various stress-responsive elements against darkness,low temperature,drought,jasmonic acid and virus stress.
