miR-34c-5p Mediated Curcumin's Effect on Reducing Lipid Accumulation in HepG2 Cells

Abstract
In order to explore curcumin's regulatory mechanism on lipid metabolism at the transcriptional level,free fatty acid-induced HepG2 cells were used as the model in this study,and cell transfection was em-ployed to adjust the intracellular miRNA levels to address whether curcumin ameliorated hepatic lipid metabolic disorders via miRNA modulation.The results demonstrated that curcumin upregulated the expression of miR-34c-5p.Overexpression of miR-34c-5p led to decreased lipid accumulation in cells,reduced content of triglyc-eride and total cholesterol,as well as downregulated expression of fatty acid and cholesterol synthesis genes,and upregulated expression of fatty acid and triglyceride catabolism genes.Further investigation revealed that some genes involved in fatty acid synthesis and elongation and glycerolipid metabolism pathways were regulated by miR-34c-5p.In conclusion,curcumin alleviated lipid metabolic disorders by increasing miR-34c-5p,thereby reducing lipid accumulation in HepG2 cells.关键词
