Optimization of Exopolysaccharide Production by Weissella confusa XG-3 in a Co-culture System and Characterization of its Properties

Abstract
In this study,a co-culture system integrating Weissella confusa XG-3 and Candida shehatae was established.The One-Factor-At-A-Time method was used to optimize exopolysaccharide production by XG3,and the properties of the produced EPS were investigated.The results demonstrated that when C.shehatae was inoculated 4 h after W.confusa XG-3 at an inoculation ratio of 1∶1,the EPS yield reached its maximum(56.82±0.12)g/L,which was 1.63 times higher than that obtained from monoculture.The optimal medium formulation for EPS production was composed of 7%sucrose,0.3%yeast extract,1.2%peptone,0.03%magnesium sulfate,0.4%ammonium citrate,0.3%dipotassium phosphate,0.05%Tween-80,5%sodium acetate,and 10%beef extract.The optimal culture conditions were an initial pH of 6.0,an incubation temperature of 30℃,and a shaking speed of 120 rpm.Based on optimization,the EPS yield of the W.confusa XG-3 strain reached(89.32±0.55)g/L,representing a 2.6-fold increase compared to before optimization.The EPS produced under co-culture exhibited excellent emulsifying activity(the emulsification indices for soybean oil and gasoline were(92.65±0.07)%and(81.08±0.09)%,respectively),a high intrinsic viscosity,and strong heavy-metal adsorption capacity.关键词
