Abstract
In order to validate the application value of DNA barcoding technology in species identification,the species of leech samples of unknown origin that were intercepted from carry-on items of inbound travelers at the Alashankou Port on the China-Kazakhstan border were identified using DNA barcoding in combination with traditional morphological method.Following preliminary morphological identification,genomic DNA was successfully extracted from the leech samples,mitochondrial COI gene sequences were obtained using PCR amplification and sequencing,and subsequently analyzed using Blast alignment against the GenBank and BOLD databases,then a phylogenetic tree was constructed using MEGA7.0 for phylogenetic analysis.The results showed that the intercepted leeches were morphologically identified as Hirudo orientalis(H.orientalis)and Hirudo lateromaculata(H.lateromaculata),respectively,their COI sequences were 100%similar with those of H.orientalis(accession No.ON098205.1)and H.lateromaculata(accession No.JN083793.1)in GeneBank,and consistent with the results searched from the BOLD database.Based on relevant literature,it was ascertained that these were firstly reported by Xinjiang ports.Exotic species such as leeches were precisely identified by the combination of DNA barcoding and morphological method,effectively supporting on-site port inspection and rapid species identification.关键词
